C4 advances cemsidomide combination testing alongside new biomarker data

C4 Therapeutics ($CCCC) presented new biomarker findings from its cemsidomide program in multiple myeloma, describing immune activation observed with cemsidomide and dexamethasone. It also reported biomarker results from the first two patients assessed in a Phase 1b combination study with elranatamab, showing expansion and activation of a specified T-cell population.

Following a six-patient safety cohort, a review committee cleared the 75-microgram dose level, permitting expansion and escalation to 100 micrograms. C4 expects results from all evaluated cohorts in mid-2027. The update combines mechanistic evidence with a dose-development milestone, rather than demonstrating improved survival or an established advantage over available therapies. The biomarker analysis covers two patients; the dose review covers six.
