Genmab reports mature survival data for petosemtamab in head and neck cancer

Genmab ($GMAB) reported median overall survival of 28.2 months with petosemtamab plus pembrolizumab in a Phase 2 study of previously untreated, recurrent or metastatic PD-L1-positive head and neck cancer. Among 43 patients evaluable for efficacy, 63% responded and 19% had complete responses. Median progression-free survival was nine months; an estimated 55% of responders maintained their response at two years.

The longer follow-up adds durability information to the experimental antibody’s development case. However, the cohort had no randomized control group, so the results cannot establish a survival advantage over existing treatment. Forty-five patients received the combination. Infusion reactions occurred in 38%, including severe events in 7%; all resolved, and no treatment-related deaths were reported. Phase 3 studies are ongoing, and petosemtamab remains investigational.
